Test Principle:

Human MUC5B(Mucin 5 Subtype B) ELISA Kit는 Sandwich-ELISA 원리를 기반으로 합니다. 키트에 포함된 마이크로 ELISA 플레이트는 Human MUC5B에 특이적인 항체가 미리 코팅되어 있습니다. 표준물질(Standards) 또는 샘플을 각 웰에 첨가하면 해당 항체와 결합합니다. 이후 Human MUC5B 특이적 biotinylated detection antibody와 Avidin-HRP (Horseradish Peroxidase) conjugate를 순차적으로 첨가하여 반응시킵니다.

반응 후 결합되지 않은 성분은 세척으로 제거됩니다. 이어서 기질(substrate) 용액을 각 웰에 첨가하면, Human MUC5B, biotinylated detection antibody, Avidin-HRP conjugate가 모두 결합된 경우에만 파란색으로 발색됩니다. 반응 정지 용액(stop solution)을 추가하면 색상이 노란색으로 변합니다.

흡광도(OD, Optical Density)는 450 nm ± 2 nm 파장에서 분광광도계를 이용해 측정하며, OD 값은 Human MUC5B 농도에 비례합니다. 표준 곡선(Standard Curve)과 비교하여 샘플 내 Human MUC5B 농도 정량 분석이 가능합니다.
